Size-Exclusion Chromatography (SEC) Columns, Waters ACQUITY UPLC Protein BEH SEC 125 Å

Size-Exclusion Chromatography (SEC) Columns, Waters ACQUITY UPLC Protein BEH SEC 125 Å

Detection of peptide aggregates, dimers and fragments by molecular size.

Size-exclusion chromatography separates molecules by size rather than by chemistry. Large species such as aggregates and dimers cannot enter the pores of the column packing and elute first; the intact peptide follows, and smaller fragments come last. The relative peak areas show how much of a sample has aggregated.

We run SEC on our UltiMate 3000 UHPLC system. Aggregation matters most for larger peptides such as GLP-1 analogues and insulin, where high-molecular-weight species can form during manufacture, shipping or storage and are not always visible in a standard reversed-phase purity test.

Applications

  • Aggregate (high-molecular-weight species) analysis of GLP-1 peptides and insulin
  • Dimer and oligomer detection in peptide and protein products
  • Monitoring stability after storage, shipping or reconstitution
  • Fragment and degradation product screening

Typical Sample Types

Peptides and proteins in solution or reconstituted from powder. Buffer and concentration are agreed per project.
